Oligonucleotide Synthesis

oligon-1.jpg

Micromon provides a high quality and reliable oligonucleotide synthesis service.

Standard DNA synthesis options are on 40 or 200 nanomole scales, using Applied Biosystems, polystyrene resin columns. The primers synthesised are suitable for direct use in most applications including DNA sequencing, standard and real-time PCR.

A key feature of Oligonucleotides made at Micromon is the extremely high level of chemical stability of the product in solution.

The Service

Standard syntheses are provided as dried, deprotected DNA aliquotted into five (5) individual tubes for the customer's convenience. This feature avoids damage to the entire oligonucleotide stock when one solution undergoes multiple freeze/thaw cycles and so prolongs the usable life of each synthesis product.

Applied Biosystems reagents and columns are used exclusively, resulting in oligonucleotides of the highest purity and quality. Synthesis couplings are monitored to ensure that the level of quality is maintained. All oligonucleotides are guaranteed.

The facility has a strong reputation for producing high quality, long oligomers designed for inclusion in synthetically constructed genes. We recommend that most oligomers, which exceed 45 bases in length, undergo stringent purification, particularly if they are to be used in the creation of synthetic genes. We offer a column-based procedure that utilises affinity chromatography technology (OPC purification). This produces a final product purity much higher than obtainable with reverse phase methods, but only marginally below that of HPLC. This purification is carried out on site at Monash and so it is an efficient time and cost effective alternative. Average yield for a 60 base oligomer is 20µg.
